生物谷報(bào)道:突觸的形成對建立神經(jīng)網(wǎng)絡(luò)十分重要,。突觸形成是一個(gè)復(fù)雜的過程。TRP通道是一類六次跨膜的非選擇性陽離子通道,。它們在進(jìn)化中高度保守,,在哺乳動(dòng)物體內(nèi)廣泛表達(dá),,參與了許多重要的生理學(xué)功能,如對溫度,、痛覺,、聽覺的感知以及受精,。TRPC通道是TRP的一個(gè)亞家族,在人的中樞神經(jīng)系統(tǒng)中表達(dá)豐富,,但其生理功能卻知之較少,。
神經(jīng)科學(xué)國家重點(diǎn)實(shí)驗(yàn)室王以政研究員領(lǐng)導(dǎo)的研究組,,發(fā)現(xiàn)TRPC6通道促進(jìn)興奮性突觸的形成,提高小鼠空間學(xué)習(xí)和記憶能力,。該研究組的博士研究生周健和杜婉璐等研究發(fā)現(xiàn)TRPC6通道在興奮性突觸,,特別是突觸后大量表達(dá),,促進(jìn)神經(jīng)元樹突棘密度的增加。樹突棘是神經(jīng)元突起上的一種微小結(jié)構(gòu),,是形成興奮性突觸的重要位點(diǎn)。他們發(fā)現(xiàn)TRPC6通道可以促進(jìn)興奮性突觸的密度,。突觸作為腦內(nèi)神經(jīng)元之間相互"對話"重要的結(jié)構(gòu),,它們數(shù)目的增加可影響學(xué)習(xí)和記憶,。為了證實(shí)這種想法,兩位研究生構(gòu)建和培育了TRPC6轉(zhuǎn)基因的小鼠,。通過行為學(xué)的實(shí)驗(yàn),發(fā)現(xiàn)該小鼠空間學(xué)習(xí)和記憶能力得到了大幅度的提高,。該項(xiàng)研究成果于2008年5月30日在線發(fā)表在神經(jīng)科學(xué)重要期刊《自然—神經(jīng)學(xué)》(Nature Neuroscience)上,。

Critical role of TRPC6 channels in the formation of excitatory synapses

The transient receptor potential canonical (TRPC) channels are Ca2+-permeable, nonselective cation channels with different biological functions, but their roles in brain are largely unknown. Here we report that TRPC6 was localized to excitatory synapses and promoted their formation via a CaMKIV-CREB–dependent pathway. TRPC6 transgenic mice showed enhancement in spine formation, and spatial learning and memory in Morris water maze. These results reveal a previously unknown role of TRPC6 in synaptic and behavioral plasticity.
