摘 要: 從油田回注水系統(tǒng)加壓泵后的管道管壁生物膜垢里分離得到的一株嗜熱,、嗜酸、異養(yǎng)型硫酸鹽還原菌, 編號(hào)為CW-04,。該菌株兼性厭氧,、革蘭氏陽性、有極生鞭毛,、直或彎曲的短桿狀,能產(chǎn)孢子, 最佳生長溫度47℃, 在20℃~63℃下培養(yǎng)基中均能使測試瓶有黑色沉淀產(chǎn)生,。從腐蝕防治的角度研究該菌株的生長曲線。主要生理生化特性包括: 生長pH范圍為3.7~10, 最佳生長pH為6~6.5, 最佳生長鹽度為0.7%,。能夠在H2+醋酸鹽,、甲酸鹽、乙酸鹽,、丙酸鹽,、乳酸鹽,、丙酮酸鹽和乙醇等電子供體中生長, 也能夠利用葡萄糖、果糖和簡單的氨基酸作為唯一碳源生長; 能夠在硫酸鈉,、硫代硫酸鈉,、亞硫酸鈉等電子受體中生長, 但是不能利用硝酸鹽和硫磺。通過16S rDNA測序以及系列比對分析: 該菌屬于細(xì)菌界(Bacteria),、厚壁菌門(Firmicutes),、梭菌綱(Clostridia)、梭菌目(Clostridiales),、蛋白胨鏈球菌科(Peptococcaceae),、脫硫腸狀菌屬(Desulfotomaculum), 與該屬中的種Desulfotomaculum aeronauticumd 相似性達(dá)到99%。

Lineages Evolution Analysis of Sulfate-reducing Bacteria in the Corrosion Biofilm

Abstract: A new thermophilic, acidityphilic, sulfate-reducing heterotrophic bacteria, strain CW-04, was isolated from biofilm dirtiness in pipeline wall after press pump of affusion system in oilfield. The bac-terium is an concurrently anaerobic, non-motile, Gram-positive, pole flagellum, spore-forming curved rod growing within a pH range of 3.7~10(optimal growth at pH 6~6.5) and temperature range of 20℃~63℃(optimal growth at 47℃), then, growth curve was studied to efficiently prevent corrosion. The optimum NaCl concentration for growth is 0.7%. A limited number of compounds are utilized as electron donors, in-cluding H2+acetate, formate, acetate, lactate, propionate, pyruvate and ethanol. Sulfate, sulfite and thiosul-fate, but not sulfer or nitrate, can be used as electron acceptors. Strain CW-04 is able to utilize glucose, fructose and simple aminophenol as sources of carbon. Analysis of the 16S rDNA sequence allowed strain CW-04(accession numbers: AY703033) to be classified as a representative of a new strain of the genus Desulfotomaculum, Desulfotomaculum aeronauticumd (Bacteria, Firmicutes, Clostridia, Clostridiales, Pep-tococcaceae) whose similarity is 99%.
