一株耐低溫纖維素酶高產(chǎn)菌株的篩選、鑒定和產(chǎn)酶的初步試驗

摘 要: 從若爾蓋高寒濕地距表層80 cm處土壤中篩選出一株纖維素酶高產(chǎn)菌株XW-1,。根據(jù)形態(tài)學(xué),、生理生化特征以及16S rDNA核酸序列分析結(jié)果表明, 該菌屬于缺陷短波單胞菌(Brevundimonas sp.)。對該菌產(chǎn)酶條件研究表明, XW-1在含0.5% CMC-Na條件下, 20°C培養(yǎng)3 d后出現(xiàn)最高酶活, 達(dá)到15.6 U/mL,。對其酶學(xué)性質(zhì)初步研究表明, 該菌株所產(chǎn)纖維素酶的最適pH為6.0, 最適反應(yīng)溫度為20°C, 15°C時相對酶活達(dá)到80%, 并且在5°C時, 相對酶活仍能保持56%,。

Screening and Identification of a Cold-adapted Cellulase-producing Bacteria

Abstract: Strain XW-1 producing extracellular cellulose was isolated from 80 cm depth under the sur-face layer at Zoige Plateau Alpine Wetland. It was identified as a Brevundimonas sp. by its morpho-logical, the physiological and biochemical properties and 16S rDNA sequence analysis. The results of cellulose producing research showed that the optimum carbon sourse was 0.5% CMC-Na, the optimum temperature was 20°C. Under this condition, the highest cellulose activity reached to 15.6 U/mL in the third day. Enzymatic properties showed that the optimum pH value is 6.0 and the optimum reaction temperature is 20°C, the relative enzyme activity reach to 80% at 15°C, moreover, at 5°C the relative enzyme activity still reach to 56%.
