2009年8月18日,,北京生命科學(xué)研究所高紹榮實(shí)驗(yàn)室在Cell Research雜志在線發(fā)表題為“Generation of induced pluripotent stem (iPS) cells from human b-thalassemia fibroblast cells” 的文章,,報(bào)道了利用b-地中海貧血成纖維細(xì)胞通過(guò)過(guò)表達(dá)四個(gè)轉(zhuǎn)錄因子獲得誘導(dǎo)多能干細(xì)胞。
β-地中海貧血癥是一種由于血紅蛋白β亞基突變而導(dǎo)致其合成降低的一種遺傳性血液疾病,。該疾病在中國(guó)南方地區(qū)較為廣泛,,目前還沒(méi)有有效的治療方法。并且,,重型β地中海貧血癥患病者由于嚴(yán)重貧血及帶來(lái)的副反應(yīng),,往往無(wú)法活到成年,。本篇文章運(yùn)用iPS技術(shù),,通過(guò)在體細(xì)胞中過(guò)表達(dá)四個(gè)轉(zhuǎn)錄因子OCT3/4,SOX2,, c-MYC和KLF4,,將病人的成纖維細(xì)胞誘導(dǎo)成為多能干細(xì)胞,所獲得的iPS細(xì)胞具有正常人ES細(xì)胞的特性,,并且可以在體外和體內(nèi)分化為不同類型的細(xì)胞包括造血細(xì)胞,,為該種疾病的治療提供了新的方法。(生物谷Bioon.com)
