靈芝作為我國傳統(tǒng)的名貴滋補(bǔ)中藥,受到國內(nèi)外的普遍關(guān)注,。從上世紀(jì)80年代起,,人們已開始進(jìn)行靈芝化學(xué)成分的研究。結(jié)果表明,,羊毛甾烷三萜化合物(lanostane)是靈芝的主要有效成分之一,,具有抗腫瘤、抗前列腺癌,、抗炎,、抑制HIV-1蛋白酶等重要生物活性,。雖然關(guān)于靈芝三萜新結(jié)構(gòu)的報(bào)道從未間斷過,但目前對結(jié)構(gòu)比較特殊的靈芝三萜的報(bào)道卻很少,。
近年來,,中國科學(xué)院昆明植物研究所資源植物化學(xué)研究組對云南省的靈芝資源進(jìn)行了收集、分離和生物活性篩選,,早在2003年就從有特色的拱狀靈芝(Ganoderma fornicatum)子實(shí)體中得到兩個(gè)新穎結(jié)構(gòu)的三萜化合物fornicatin A, B,,隨后深入研究該子實(shí)體,又發(fā)現(xiàn)了fornicatin C,,這些化合物都是拱狀靈芝中含量很高的特色三萜化合物,。
最近,該組的碩士研究生王翠芳等人在導(dǎo)師邱明華研究員的指導(dǎo)下,,對購買自楚雄南華野生菌市場的紫芝(Ganoderma sinensis)進(jìn)行了研究,。通過提取、分離,、結(jié)構(gòu)鑒定,,從該紫芝中獲得了3個(gè)C-1與C-11位直接相連形成五個(gè)環(huán)的靈芝三萜化合物methyl ganosinensate A, ganosinensic A和ganosinensic B,。這3個(gè)化合物是一類新穎的五環(huán)骨架體系,,這在三萜中是首次出現(xiàn)的。依據(jù)這些化合物的結(jié)構(gòu),,研究人員提出了methyl ganosinensate A一個(gè)可能的生物合成路線,。這一發(fā)現(xiàn)進(jìn)一步充實(shí)了天然二次代謝產(chǎn)物的三萜結(jié)構(gòu)類型。

Methyl ganosinensate A (1), ganosinensic acid A (1a), and ganosinensic acid B (2), three new triterpenoids with an unusual four-membered ring skeleton produced by a bond across C-1 to C-11, were isolated from the fruiting body of Ganoderma sinense. Their structures were established on the basis of extensive spectroscopic methods, including 1D and 2D NMR techniques, and methyl ganosinensate A was confirmed by X-ray crystallographic analysis.
