禾本科植物內(nèi)生真菌研究5. 中國產(chǎn)部分epichloë內(nèi)生真菌的遺傳多樣性

摘 要:Epichlo?內(nèi)生真菌(epichlo? endophytes)是禾本科植物的共生真菌,,開展其多樣性研究對菌株的生物學(xué)特性和進化規(guī)律的了解以及開發(fā)利用都有著十分重要的意義,。本研究篩選了12條隨機引物對24株epichlo?內(nèi)生真菌進行RAPD擴增,探討真菌種類,、宿主種類,、采集地和菌株間的遺傳多樣性。結(jié)果表明,,宿主為拂子茅Calamagrostis sp.、雀麥Bromus sp.,、披堿草Elymus sp.,、小穎羊茅Festuca parvigluma Steud.的菌株分別獨立聚為一個分枝;而宿主為鵝觀草Roegneria spp.的Neotyphodium屬菌株聚類情況較為復(fù)雜,;宿主為早熟禾Poa spp.的菌株分別在三個不同的分枝,,表現(xiàn)出豐富的遺傳多樣性。以上結(jié)果反應(yīng)了我國具有豐富的epichlo?內(nèi)生真菌資源,,它們和其宿主之間有著比較明顯的相互關(guān)系,。本研究還顯示,采集于南京的拂子茅與其內(nèi)生真菌共生可能發(fā)生在較久遠(yuǎn)的年代,。

Grass endophyte researches 5. Genetic diversity in some Chinese epichloë endophytes

Abstract: Mutualistic associations between epichlo? endophytes and gramineous plants are receiving more and more attention. Investigations of the endophytes' diversity are facilitative to understand their biological properties and evolutionary relationships. Random amplified polymorphic DNA (RAPD) markers were used to investigate genetic diversity and evolutionary characteristics of 24 endophytic fungal isolates. Isolates from Calamagrostis sp., Bromus sp., Elymus sp. and Festuca parvigluma Steud. significantly clustered respectively; clusters for Neotyphodium isolates obtained from Roegeria spp. showed comparatively complicated, while isolates obtained from Poa spp. tended to be divided into three groups. It could be concluded that the genetic relationship among the epichlo? endophytes native to China is quite diverse. The results also indicated the mutualistic relationship between Calamagrostis sp. collected from Nanjing and its endophyte could be traced back to ancient time.
