生物谷從武漢大學(xué)醫(yī)學(xué)院免疫學(xué)系獲悉,,該校譚錦泉教授及其助手張秋萍副教授等人率先“捕獲”了T淋巴細(xì)胞性白血病的癌細(xì)胞,,抵抗各類抗癌藥物的首號“元兇”——一種名為Livin的蛋白質(zhì)。有關(guān)該研究的論文,,已經(jīng)發(fā)表在10月15日出版的國際上最具權(quán)威的腫瘤研究刊物“Cancer Research”《美國腫瘤研究》刊物上,。
譚錦泉教授和課題組專家們曾在世界上首次了發(fā)現(xiàn)T淋巴細(xì)胞性白血病的癌細(xì)胞CCR9表達(dá)異常增高時,對心,、肝,、腎、腦等人體主要器官有強(qiáng)致病性的成果,。
譚錦泉教授等在研究中,,發(fā)現(xiàn)白血病癌細(xì)胞上異常活躍的趨化因子受體CCR9,,可以在癌細(xì)胞體內(nèi)形成了一條十分“詭秘”的“通道”,這條“通道”上第三站的“神秘人物”每次“表現(xiàn)”活躍時,,該癌細(xì)胞就體現(xiàn)出強(qiáng)大的生命力,,可以十分順利地“逃脫”或“躲避”多種抗癌藥物的打擊,而使該癌細(xì)胞“程序化死亡”的機(jī)率大大降低。他們經(jīng)過無數(shù)次實驗,,最終發(fā)現(xiàn)這個“神秘人物”就是一種叫“Livin”的蛋白質(zhì),,并且“幫兇”正是異常活躍的CCR9,。
據(jù)譚錦泉教授介紹,,這種名為“Livin”的蛋白質(zhì)是該癌細(xì)胞中抵抗藥物作用最頑固的,所以稱之為頭號“元兇”,。他希望,,在今后的研究中,還能“捕獲”更多的“元兇”,,真正使抗癌藥物有的放矢,、藥到病除,真正讓人類遠(yuǎn)離白血病的折磨,。
從事該研究的張秋萍副教授介紹,,白血病是人體造血系統(tǒng)的一種高度異質(zhì)性疾病,其一個重要的病理改變就是發(fā)生病變的細(xì)胞向體內(nèi)肝,、心,、腦、腎,、脾等重要器官游走浸潤,,引起這些器官不可逆的損害,從而直接威脅患者生命,。而T淋巴細(xì)胞性白血病是根據(jù)白血病免疫學(xué)特征分類中的一種,。
譚錦泉等通過對大量白血病人血液標(biāo)本進(jìn)行研究后發(fā)現(xiàn),T淋巴細(xì)胞性白血病患者體內(nèi)T淋巴細(xì)胞上的趨化因子受體CCR9均有異常增高,,從而誘導(dǎo)了病變細(xì)胞向各重要臟器浸潤,,進(jìn)而造成對白血病患者的器官損害。
研究者們還發(fā)現(xiàn),,對T淋巴細(xì)胞性白血病患者的T淋巴細(xì)胞聯(lián)合應(yīng)用白介素-2和白介素-4,,可以明顯抑制趨化因子受體CCR9在T淋巴細(xì)胞上的表達(dá),從而大大降低這類患者器官受損的可能,。
據(jù)介紹,,與目前普遍使用的放療、化療相比,,這種生物療法更為高效,、無毒。我國白血病的自然發(fā)病率為十萬分之三,,如能在臨床上推廣使用這一方法,,無疑對白血病患者是一個巨大的福音,。
