阿爾茨海默氏?。ɡ夏臧V呆癥)是發(fā)病率很高的一種神經(jīng)退行性疾病,,造成嚴(yán)重的經(jīng)濟(jì)、社會(huì)和家庭負(fù)擔(dān),。Beta淀粉樣蛋白質(zhì)(Aβ)的聚集體(球形寡聚體,,纖維,斑塊等)對(duì)腦神經(jīng)細(xì)胞產(chǎn)生損傷,,甚至誘發(fā)神經(jīng)細(xì)胞死亡,,最后導(dǎo)致癡呆發(fā)生。理解A?的聚集機(jī)理以及對(duì)聚集行為進(jìn)行有效的調(diào)控是深入了解老年癡呆癥發(fā)病機(jī)理以及早期診斷,、干預(yù)和治療的關(guān)鍵,。
國(guó)家納米科學(xué)中心研究人員近期在研究Aβ等淀粉樣蛋白的聚集機(jī)理以及利用類(lèi)分子伴侶的調(diào)節(jié)效應(yīng)對(duì)淀粉樣蛋白的聚集行為進(jìn)行有效調(diào)控方面取得新進(jìn)展。他們利用掃描隧道顯微技術(shù)研究了阿爾茨海默氏病相關(guān)的多肽Aβ42(J. Mol. Biol. 2009, 388, 894) 以及II型糖尿病相關(guān)的胰淀素(Amylin)(J. Struct. Biol., 2009, 167, 209) 的組裝機(jī)理,,對(duì)于具有beta片層結(jié)構(gòu)的多肽組裝結(jié)構(gòu)的解析和組裝機(jī)理的解釋提供了新的思路,。在此工作基礎(chǔ)上,他們報(bào)道了一種利用分子調(diào)節(jié)劑有效調(diào)控淀粉樣蛋白聚集的新方法,,利用類(lèi)分子伴侶的調(diào)節(jié)效應(yīng)不僅可以在分子水平上有效調(diào)控多肽的二維組裝特性,,而且可以調(diào)控多肽的三維聚集行為。此項(xiàng)研究從分子水平上揭示了多肽與分子調(diào)節(jié)劑相互作用的機(jī)理,,為與淀粉樣蛋白聚集有關(guān)的疾病的預(yù)防,、藥物設(shè)計(jì)和治療提供了潛在的可行途徑。有關(guān)研究成果已經(jīng)發(fā)表在近期的《納米快報(bào)》(Nano Lett. 2009, 9(12), 4066-4072)上,。

Chaperon-Mediated Single Molecular Approach Toward Modulating Aβ Peptide Aggregation

We report here a single molecular approach using chaperone-like molecular modulators for modulating the aggregation behavior of a vital analogue of beta-amyloid peptide (Aβ) by using scanning tunneling microscopy. The molecular structures of the β-sheets for Aβ33?42 peptide are revealed, which are keen to the aggregation of Aβ42 relating to Alzheimer’s disease. It was identified that the introduction of chaperone-like modulators could regulate the assembling behavior of the peptide at molecular level. Furthermore, the modulators could also significantly accelerate the aggregation of the peptide in aqueous solution as revealed by light scattering studies. These observations of the molecular modulator effect in peptide assemblies could provide a novel approach toward modulating Aβ peptide aggregations.
