生物谷報(bào)道:在過(guò)去的十年里,,芯片技術(shù)已經(jīng)在基因表達(dá)研究中起到了舉足輕重的作用,,帶來(lái)了深廣的影響?;蛐酒玫胶A繑?shù)據(jù),,但這些海量數(shù)據(jù)是否準(zhǔn)確?如何最大限度提高芯片數(shù)據(jù)的準(zhǔn)確性,,是科學(xué)家們最頭疼的問(wèn)題,。尤其是但是隨著利用不同芯片平臺(tái)獲得相異甚至大相徑庭的結(jié)果逐漸增多,,許多人產(chǎn)生了對(duì)這一技術(shù)可靠性的質(zhì)疑爭(zhēng)論。芯片質(zhì)量控制(MicroArray Quality Control,,MAQC)計(jì)劃因而得以誕生,,作為一個(gè)標(biāo)準(zhǔn)將對(duì)相關(guān)數(shù)據(jù)分析和爭(zhēng)論進(jìn)行評(píng)定。
最新著名的基因芯片專(zhuān)家,,華人學(xué)者石樂(lè)明(音譯)以同一期Nature biotech 上發(fā)表了5篇文章系統(tǒng)闡述有關(guān)基因芯片質(zhì)量控制的問(wèn)題解決。
外源RNA對(duì)照(External RNA controls,,ERCs)是MAQC的一個(gè)重要部分,,對(duì)于芯片分析性能測(cè)評(píng)也是非常重要的一個(gè)指標(biāo),但是在各研究團(tuán)體中并未能得到充分利用,。在這一研究報(bào)告中,,研究人員對(duì)兩類(lèi)ERCs進(jìn)行了評(píng)估:其中一類(lèi)在樣品擴(kuò)增和標(biāo)記之間加入到總RNA中,另外一類(lèi)怎是在雜交之前加入到copyRNAs (cRNAs)中,,通過(guò)多商業(yè)芯片平臺(tái)(multiple commercial microarray platforms獲得了ERC濃度反應(yīng)曲線(xiàn)(concentration-response curves),,辨認(rèn)出在分析過(guò)程中出現(xiàn)問(wèn)題的分析以及潛在的變量來(lái)源,除此之外研究人員也對(duì)不同的ERC類(lèi)型的行為進(jìn)行了檢測(cè),。這些ERC評(píng)估將為未來(lái)的多平臺(tái)分析性能評(píng)估中ERC的更好使用提供了一個(gè)基礎(chǔ),。在這一研究中,浙江大學(xué)部分研究工作者參與了這一計(jì)劃,。

Dr. Shi co-authored about 100 peer-reviewed publications in chemoinformatics, bioinformatics, and data mining.
