近日,,中國科學(xué)院植物研究所研究員田世平帶領(lǐng)的研究小組,利用雙向凝膠電泳結(jié)合MALDI-TOF/TOF質(zhì)譜分析技術(shù),,發(fā)現(xiàn)灰霉病菌在不同環(huán)境pH值條件下胞外分泌組有顯著差異,,共鑒定出47個(gè)差異蛋白點(diǎn),對應(yīng)21個(gè)不同的胞外蛋白,。相關(guān)研究成果近期在線發(fā)表在國際著名學(xué)術(shù)期刊Journal of Proteome Research上,。
同時(shí),研究發(fā)現(xiàn)在pH4條件下,,灰霉病菌傾向于分泌更多的蛋白酶類,,而在pH6條件下與細(xì)胞壁降解相關(guān)的酶類被誘導(dǎo)表達(dá)。實(shí)時(shí)定量PCR的分析結(jié)果表明,,絕大部分差異蛋白的表達(dá)調(diào)控發(fā)生在轉(zhuǎn)錄水平上,。
灰霉病菌能引起世界上200余種植物發(fā)生病害,,每年造成巨大的經(jīng)濟(jì)損失,。該病菌不僅寄主范圍廣,而且能侵染寄主植物的果實(shí),、花器,、莖稈、葉片等不同組織,。研究灰霉病菌的致病機(jī)制,,對預(yù)防和控制灰霉病的發(fā)生具有重要意義。
上述研究結(jié)果表明,,灰霉病菌可以通過調(diào)節(jié)分泌蛋白表達(dá)譜來響應(yīng)環(huán)境pH值(包括寄主組織pH值)的變化,,這為揭示灰霉病菌在不同寄主和組織上復(fù)雜的侵染機(jī)制提供了重要依據(jù),。(生物谷Bioon.com)

Exploring Pathogenic Mechanisms of Botrytis cinerea Secretome under Different Ambient pH Based on Comparative Proteomic Analysis

Botrytis cinerea causes gray mold rot on over 200 plant species worldwide, resulting in great economic loss every year. Cooperation of proteins secreted by B. cinerea plays an important role in its successful infection to host plants. The ambient pH, as one of the most important environmental parameters, can regulate expression of secreted proteins in various fungal pathogens. In the present study, we mainly investigated the effect of ambient pH on secretome of B. cinerea strain B05.10 with a comparative proteomic method based on 2-DE. Distinct differences in secretome of B. cinerea were found between pH 4 and 6 treatments, and 47 differential spots, corresponding to 21 unique proteins, were identified using MALDI-TOF/TOF. At pH 4, more proteins related to proteolysis were induced, whereas most of up-accumulated proteins were cell wall degrading enzymes at pH 6.
